Why a keystone?

I like an explanation with a small, essential piece. I like it even more when the experiments complicate that explanation.

An editorial site about kisspeptin

Keystone Peptides grew from my fascination with published research: a biological signal whose importance becomes visible when its reception goes wrong. The human genetics of GPR54 and pubertal failure gives that fascination a concrete starting point. Kisspeptin deserves the attention because the relationship is specific enough to investigate.

The name is an editorial title. There is no medical business here, and nothing on these pages is medical advice. An assessment of symptoms or a treatment decision is beyond this site's purpose. Those decisions require the circumstances of an actual person and a qualified medical professional; an absorbing paper cannot provide that relationship.

Enthusiasms and reservations

I favor awkward experiments that improve an explanation. The loss of hormonal response with repeated kisspeptin administration is a favorite example. It leaves the importance of the signal intact while complicating the idea that more stimulation will reliably help.

I am enthusiastic about the egg-maturation trial in women undergoing IVF. Its appeal is the precise task. I am also interested in the food-intake experiment that found an LH response without a change in hunger or intake. That finding narrows the story in a useful way. A molecule can deserve sustained curiosity without having every hoped-for effect.

Where the metaphor ends

I distrust the point at which a pleasing comparison starts making decisions for the biology. A keystone suggests something central and stable. The receptor pharmacology introduces changing responsiveness, desensitization and recovery. The living system is more interesting than the image.

That tension is the reason this site exists. I want the pivotal role and the inconvenient behavior in the same account. The discovery history, rhythm piece, IVF report and essay on desire approach that interest from different directions. The question connecting them is how a signal acquires its importance in a particular setting, and how much of that importance survives an attempt to put it to work.
